Pematangan gonad, pemijahan, penetasan telur dan perawatan larva ikan gabus (Channa striata)

 Abstrak

Ikan gabus (Channa striata) adalah salah satu jenis ikan perairan umum tawar dengan nilai ekonomi tinggi. Selain sebagai bahan pangan (konsumsi), ikan ini juga sebagai sumber albumin untuk industri farmasi. Untuk memenuhi kebutuhan masyarakat dan industri terhadap ikan ini masih mengandalkan hasil tangkapan dari alam. Pembudidayaan ikan gabus di Indonesia belum ada karena belum tersedia paket teknologinya. Penelitian untuk membenihkan ikan gabus di Indonesia sudah mulai dilakukan. Pematangan gonad menggunaan hormon human chorionic gonadotropin dosis 300 internasional unit, menghasilkan gonadosomatic index sebesar 2,87-11,54%. Pemijahan semi alami dapat mempercepat waktu latensi, suhu inkubasi 28oC daya tetas telur sebesar 86,33%, padat tebar larva pada media pemeliharaan 2 ekor/L menghasilkan survival rate sebesar 63,83%.

DNA authentication of asian redtail catfish Hemibagrus nemurus from musi and penukal river, south sumatra indonesia

Abstrak

Asian Redtail Catfish (Hemibagrus nemurus) is highly economic important catfish in South Sumatra. Investigating DNA authentication is of importance for species conservation and breeding selection. Cytochrome C Oxidase subunit I is one of mitochondrial DNA markers used for species barcoding in freshwater, brackishwater and marine fish. This research aims to explore the use of COI gene for species barcoding, constructing phylogenetic tree of Asian Redtail Catfish (ARC). The methods used in the research consisted of DNA extraction, PCR (Polymerase Chain Reaction) amplification and sequencing mtDNA COI gene of ARC obtained from Musi and Penukal Rivers (South Sumatra). A 572 and 596 base pairs of partial coding sequences were obtained representing each river. Phylogenetic analyses indicated that ARC was at the same cluster from Bagridae family, and different cluster from others catfish, Pangasius sp and Oreochomis niloticus. Further study using more species of Bagridae and habitat are needed to investigate the diversity of DNA from South Sumatra water resources

DNA barcodes and phylogenetic of striped snakehead and ocellated snakehead fish from South Sumatra, Indonesia.

Abstract.

This research aimed to identify the sequences of cytochrome c oxidase subunit I gene mitochondrial DNA (COI mtDNA), to construct a phylogenetic tree of striped snakehead (Channa striata) and ocellated snakehead (Channa pleuropthalma), and to measure water quality of Kelekar River, Indralaya, Ogan Ilir District and Danau Burung Besar River, Penukal Abab Lematang Ilir (PALI) District in South Sumatra, Indonesia. The research procedures consisted of DNA isolation, amplification by PCR (Polymerase Chain Reaction) and sequencing of fragment COI mtDNA. The length of nucleotide was 604 bp for striped snakehead and 587-604 bp for ocellated snakehead. Optimum annealing temperature was 500C for 15 seconds with 30 cycles. The result of BLAST analysis showed that striped snakehead from Kelekar and Danau Burung Besar River had 100% identity to striped snakehead from Java-Bali and furthest (97%) with striped snakehead from India. Ocellated snakehead had 100% similarity with the same species from Musi Banyuasin and Banjarmasin; and furthest (83%) with Channa limbata from Myanmar. Water quality in Kelekar River were temperature 31-31.6 0C, pH 4.76-4.96, dissolved oxygen 2.7-3.0 mg/L, ammonia <0.009 mg/L, total alkalinity 20 mg/L, and turbidity 62.5-63 cm. Meanwhile in Danau Burung Besar River showed temperature (29.3-30.7 0C), pH (3.6-6.7), dissolved oxygen (1.31-3.76 mg/L), ammonia (0.17-0.20 mg/L), and turbidity (50-90 cm).
