Biodiversity of Freshwater Fish in Kelekar Floodplain Ogan Ilir Regency in Indonesia

 

Abstract

The purpose of this study is to investigate fish biodiversity in the Kelekar floodplain. The study is explorative, with the determination of observation stations and with purposive sampling methods. Fishes were captured approximately 1.509 individuals consisting of 17 families and 24 species. The Shannon-Weiner diversity index was 2.394; 2.691; and 2.183 for station 1, 2, and 3, respectively. The Evenness index was 0.764; 0.871; and 0.806 for station 1, 2, and 3, respectively, meanwhile the highest value of Simpson’s dominance index was 0.045. The biodiversity index of the three stations was in the medium category.

Characterization of genes encoding follicle-stimulating hormone β-subunit (fsh-β) and luteinizing hormone β-subunit (lh-β) from Indonesian leaffish Pristolepis grootii (Bleeker, 1852)

ABSTRACT

The Indonesian leaffish Pristolepis grootii (Bleeker, 1852) is an indigenous fish species. The purpose of this study was to analyze the characteristics of the β-subunit of follicle-stimulating hormone (FSH-β) and the β-subunit of luteinizing hormone (LH-β) of P. grotii. Complementary DNA (cDNA) was synthesized from mRNA that was extracted from P. grootii pituitary glands. Twelve fish were reared in two aquaria and injected with luteinizing hormone-releasing hormone analog (LHRHa) for the analysis of FSH-β and LH-β gene expression. The results showed that P. grootii FSH-β was 227 bp in length, consists of 93 amino acid residues, while LH-β was 300 bp in length and consists of 100 amino acid residues. P. grootii FSH revealed the highest similarity (91%) with climbing perch Anabas testudineus, while LH-β with Betta splendens (90%). Two putative N-glycosylation sites in FSH-β and one site in LH-β were conserved in P. grootii. The expression of FSH-β and LH-β in fish injected with LHRHa was higher than that of LHRHa not injected fish. The ovary of P. grootii had various sizes of oocytes. The injection of LHRHa affected the development of oocytes; there was a trend for higher doses LHRHa to develop oocytes faster. Thus, it is most likely that the isolated genes are FSH-β and LH-β of P. grootii.

Ovary development, FSH and LH genes expression of Indonesian leaffish (Pristolepis grootii) (Bleeker, 1852), injected with luteinizing hormone-releasing hormone analog

 ABSTRACT 

Indonesian leaffish, Pristolepis grootii (Bleeker, 1852), is an undomesticated freshwater fish species native to the rivers, flooded swamps, and tributaries of Indonesia. The fish is mainly captured for consumption. In order to prevent its extinction and supply its growing demands, the artificial breeding of the fish should be developed. The purpose of this study was to determine the optimum dose of luteinizing hormone-releasing hormone analog (LHRHa) for stimulating the female P. grootii gonadal development at a dosage of 0, 1, 10, and 50 µg kg-1 of fish. Female fish (20.0 ± 0.6 g) were adapted for 30 days in the rearing environment and then separated into 12 aquariums with six fish per aquarium. Fish were then reared for another 21 days and fed with Tubifex sp. The LHRHa injection was conducted twice on day-7 and 14. Fish bodyweight, gonadosomatic index, gonad histology, blood estradiol-17β, and FSH-β and LH-β gene expression were evaluated at day 0, 7, 14, and 21. The results showed that the injection of the LHRHa hormone stimulated the development of fish gonads and was better achieved with a higher concentration of LHRHa. The best treatment was observed by the administration of 50 µg kg-1 of LHRHa that produced the fastest development among all treatments. This study demonstrated that the LHRHa induction could potentially stimulate the gonadal development of the newly domesticated fish. To our knowledge, this is the first study that reported the success of the induction of female gonad development in the Indonesian leaffish P. grooti.
