PERTUMBUHAN DAN KELANGSUNGAN HIDUP LARVA TAMBAKANG (Helostoma temmincki) DIRENDAM DALAM HORMON TIROKSIN DENGAN LAMA PERENDAMAN BERBEDA

ABSTRACT

The aim of this research were to determine of the best emmersion duration to accelerate growth of kissing gouramy. The research was conducted on juli until august 2014 in Fish Breeding unit Batanghari Sembilan Indralaya, Kabupaten Ogan Ilir. This research used Completely Randomized Design (CDR), with 4 treatment 0, 12, 24, 36 hour of immersion, with 4 replications. The parameters observed were length (mm), weight (mg), survival rate (%) and water quality. The results of this research shown the highest growth of kissing gouramy larvae with immersion duration is 24 hours with the average value of 23,93 mm of length and increase in weight of  265,31 mg. The best survival rate is 24 hour duration of immersion with average value 64,29%. The average value of water quality 25,5-30,1oC for temperatur, 6,5-7,0 for pH, 4,45-4,97 mg.L-1 for dissolved oxygen and 0,05-0,08 mg.L1 for ammonia.

DNA Authentication of Asian Redtail Catfish Hemibagrus nemurus from Musi and Penukal River, South Sumatra Indonesia

Abstract

Asian Redtail Catfish (Hemibagrus nemurus) is highly economic important catfish in South Sumatra. Investigating DNA authentication is of importance for species conservation and breeding selection. Cytochrome C Oxidase subunit I is one of mitochondrial DNA markers used for species barcoding in freshwater, brackishwater and marine fish. This research aims to explore the use of COI gene for species barcoding, constructing phylogenetic tree of Asian Redtail Catfish (ARC). The methods used in the research consisted of DNA extraction, PCR (Polymerase Chain Reaction) amplification and sequencing mtDNA COI gene of ARC obtained from Musi and Penukal Rivers (South Sumatra). A 572 and 596 base pairs of partial coding sequences were obtained representing each river. Phylogenetic analyses indicated that ARC was at the same cluster from Bagridae family, and different cluster from others catfish, Pangasius sp and Oreochomis niloticus. Further study using more species of Bagridae and habitat are needed to investigate the diversity of DNA from South Sumatra water resources

PERBEDAAN JUMLAH KROMOSOM IKAN TOMAN (Channa micropeltes) DENGAN IKAN SERANDANG (Channa pleuropthalmus)

ABSTRACT
The aims of this research was to comparing the chromosomes number from Channidae family. They are giant snakehead (C. micropeltes) and ocellated snakehead (C. pleuropthalmus). The research was conducted in December 2013-January 2014 in the aquaculture Laboratory, Aquaculture Study Program, Agriculture Faculty, Sriwijaya University for acclimatization of fish, Animals Fisiology Laboratory, Biology Sriwijaya University for made preparations and Laboratory  genetic and fish reproductcy Departement of aquaculture, Maritime Affairs and Fishery Faculty Bogor Agriculture Institute for chromosome analysis. Weight of the fish used in this study was 500-600 gram. Analysis performed on chromosome preparations of both fish used squash method with modifications and stain with Giemsa. Based on the results of the two types of fish that Channa genus that have different numbers of chromosomes where giant snakehead has 50 of chromosome number with range 2n=40-50 and ocellated snakehead has 46 of chromosome number with range 2n=43-46.

ABSTRACT

The aims of this research were to determining the right changed periode of Artemia sp., Moina sp., and artificial feed as fed of climbing perch (Anabas testudineus) larvae to growth and survival rate. The research used completely randomized design with three treatments and three replications. The treatment was difference changed periode of Artemia sp., Moina sp., and artificial feed as fed of climbing perch (Anabas testudineus) larvae. Artemia sp. were fed at 3–15 days after hatching,  Moina sp. were fed at 14 – 24 days after hatching and artificial feed at 21–33 days after hatching (I: 3-15, II: 14-24, III: 21-33) as treatment A;  Artemia sp. were fed at  3–11 days after hatching,  Moina sp. were fed at 10 – 20 days after hatching, and artificial feed at 17–33 days after hatching (I: 3-11, II: 10-20, III:17-33) as treatment B; Artemia sp. were fed at 3–7 days after hatching,  Moina sp. were fed at 6-18 days after hatching and artificial feed at 15–33 days after heatching ( I: 3-7, II: 6-18, III: 15-33) as treatment C. Fish were fed three times a day at 8 am, 10.30 am, 1 pm, 3.30 pm, 6 pm using at satiation method. Based on Analysis of Variance (ANOVA), the results showed significant difference effect in specific weight growth rate but no significant difference effect in specific lenght growth rate. Result of Honestly Significance Difference test showed the highest specific weight growth rate was treatment C, but no significant difference with treatment B. The best treatment for survival rate was treatment A  which significant difference with treatment C but no significant difference with treatment B. As long as research, water quality was in optimum range for  climbing perch (Anabas testudineus) larvae  rearing. Temperature was  27-28 oC , DO 3.38-5.06 mg.L-1 , pH 4.4-6.6, ammonia 0.002-0.008 mg.L-1.

ABSTRACT

The problem of development of climbing perch culture was low in growth rate. The effort to increase growth rate of climbing perch is hormonal treatment, including the hormone thyroxine. The aim of this research was to determine the effective consentration and duration of immertion of thyroxine hormone to increase yolk sack absorbtion and growth rate. This research held in August to September 2014 in Laboratorium Dasar Perikanan, Aquculture Study Departement, Faculty of Agriculture, Sriwijaya University, Indralaya. This research using factorial completely rondomezed design with two factors, the first was thyroxine consentration with immersion media (K1=0.01 mg.L-1, K2=0.05 mg.L-1, K3=0.1 mg.L-1) and the second was the long immersion (W1=24 hours, W2=48 hours, W3=72 hours) with three replication. The parameters that observed were yolk sack absorption, growth, survival rate and water quality. The result showed that the highest yolk sack absorbtion was K2 (0.05 mg.L-1) and K3 (0.1 mg.L-1). The highest growth was K3W2 (0.1 mg.L-1 of consentration and immersion for 48 hours) with the weight was 316.1 mg and length was 26.56 mm. The best survival rate was 88.67% (K1W1). The value of water quality during larva rearing were temperature 26-28 oC, pH 5.6-6.5, dissolved oxygen 4.435.93 mg.L-1 and ammonia 0.00-0.03 mg.L-1

ABSTRACT

The objective of this study is to know the best synthetic gonadotrophine hormon doses to stimulate the breeding of snakehead through observing the latent time, the number of eggs, and the percentage of fertilized eggs and the hatching percent of snakehead. This experiment was conducted in (UPR) Batanghari Sembilan in North Indralaya sub-district in Ogan Ilir regency on January until February 2015. The design of this study was completely randomized design which having three different treatments of different doses of hormone given. The doses treatments were P1 = 0.2 ml/kg fish, P2 = 0.4 ml/kg fish and P3 = 0.6 ml/kg fish with each male and female parent of fish was injected three treatment trhee times for each treatments. The result of this experiment showed that utilization of synthetic gonadotrophine hormone with different doses had significant different effect to hatching percentage did not significantly different (P<5%) to latent time, the amount of eggs, and fertilized eggs percentage of snakehead. In this experiment, the treatment P1 was the best terms of four parameters which were the latent time (27.70 hours), the amount of eggs (6,668 eggs), the fertilized eggs percentage (99.75 %), and hatching percentage (78.47 %). Futhermore, the value range of water quality during the experiment were temperature 28320C, pH 3.7-7.0 and dissolved oxygen 3.08-5.76 ppm.

ABSTRACT

The aim of this research was to know the different of body weight ratio on spawning latency period, total eggs, percentage of fertilized eggs and percentage of hatched eggs. The research was conducted from March to April 2015 in Laboratorium Budidaya Perairan, Agriculture Faculty, Sriwijaya University and Fish Breeding Unit Batanghari Sembilan, Indralaya, Ogan Ilir. The research used experimental methode and it was arranged in a completely randomize design with four treatments and three replications. The pituitary extract induction with donor and recipient body weight ratio 1:1 (P1), 2:1 (P2), 3:1 (P3) and sintetic hormone induction (KP). The parameters observed were spawning latency period, total eggs, percentage of fertilized eggs and percentage of hatched eggs. The results showed that percentage of fertilized eggs 98.23-99.54% and percentage of hatched eggs 51.66-64.33%. The fastest of spawning latency period were showed by P3 20.47 hours while the best of total eggs were showed by P3 6,112 eggs.

ABSTRACT

The aims of this research was to know the effect of fed with addition of probiotic to bacteria population, and water quality of media maintenance, histology of snakehead (Channa striata) fry. This research used randomized completely design (RCD) with six treatments and three repeat with treatments code (D). treatment tested were D0 (control without probiotic), D1 (2.5 ml.kg-1 feed), D2 (5 ml.kg-1 feed), D3 (7.5 ml.kg-1 feed), D4 (10 ml.kg-1 feed), D5 (12.5 ml.kg-1 feed). The research was conducted November to December, 2012 at the Laboratorium Dasar perikanan, Agriculture Faculty Sriwijaya University, Indralaya. Population bacteria, water quality and histology were analyzed descriptively. The result research was conducted that addition probiotic with a cocentration of 10 ml.kg-1 feed in snakehead medium give good influence to survival rate (63.33 %). Histology of intestine were enteritis, loss of epithelial tunica serosa, and histology of lever were congestion, necrosis, hemorrhagic.

ABSTRACT

The aims of this research was to determine the effect of probiotics in population bacteria intestinal, histology, survival rate and growth of snakehead. The research was carried out for 30 days of maintenance in a trap as a media container maintenance. This research used the method of completely randomized design with six treatments and repeated in three times. The treatments are W0 (control or without probiotic), W1 (2.5 µl.l-1  week-1), W2 (5 µl.l-1 week-1), W3 (7.5 µl.l-1 week-1), W4 (10 µl.l-1 week-1) dan W5 (12.5 µl.l-1 week-1). The result of the research showed that addition of probiotic in media of snakehead (C. striata) obtained the best dose is 10 µl.l-1 week-1, which can have to reduce the population of phatogenic bacteria in intestinal and had a good effect for survival rate (93.33%). As for the histopathologic known that probiotics do not give significantly different effect.

PERTUMBUHAN POPULASI Daphnia sp. DENGAN PEMBERIAN LARUTAN  KULIT SINGKONG TERFERMENTASI

ABSTRACT
Fermented cassava peels is one of the potential nutrient for growth of Daphnia sp population. This research aim was to know the influence of fermented cassava peels with yeast as source nutrition food to population growth of Daphnia sp.. This research was conducted in May 2016 Laboratory of Aquaculture, Aquaculture Program Study Sriwijaya University. This study used a complete randomized design (CRD) with four treatments and three replications i.e Daphnia sp. fed with fermented cassava peels of 0.05, 0.10, 0.15 and 0.2 g.mL-1. The data collected is a test of proximate peels before and after fermented cassava, population densities of Daphnia sp., the growth rate of population Daphnia sp., time achievement of Daphnia sp. population peak and physical chemistry of water. Research results show that fermentation by yeast can improve the nutritional value of cassava peels as a protein in by 0,45 and lower carbohidrate of 6,79, ash content  1,31, 1,04 fat and HCN 3  Provision of fermented cassava peel solution of 0.20 g.mL-1 produces a peak population of 494 ind.L-1 to 7.3 days and the rate of population growth as much as 43.91% .days-1. Physical chemistry of water during the study are in the optimum range for growth of Daphnia sp.

PERSENTASE PENETASAN TELUR IKAN GABUS (Channa striata) PADA PH AIR YANG BERBEDA 

ABSTRACT 
The success in egg hatching is determined by internal and external factors. One of external factors is the acid level. The use of pH in egg hatching is to stimulate chorionase enzyme that can make chorion become soft. The purpose of this research is to determine the best pH value for hatching of snakehead eggs. This research had been conducted in Laboratorium Dasar Perikanan, Departement of Aquaculture, Agriculture Faculty, Sriwijaya University on January until February 2016. The research method used a completely randomized design with five treatments and three replications. The treatment were P1 (pH 5±0.2), P2 (pH 6±0.2), P3 (pH 7±0.2), P4 (pH 8±0.2) dan P5 (pH 9±0.2). The results showed that different values of water pH in snakehead hatching gave significant effect on hatching percentage, incubation time and survival rate of larvae but did not indicate  significant effect on percentage of abnormal larvae. The highest hatching percentage was in treatment P5 (90.75%), the fastest incubation time was in treatment P4 (20.00 hours), the highest survival rate of larvae was in treatment P2 (85.31%) and the highest percentage of abnormal larvae was in treatment P1 (1.67%). During the research, water quality were in tolerance range for hatching and snakehead larvae rearing where DO (5.27-6.01 mg/l), ammonia (0.00-0.29 mg/l), and alkalinity (26-106 mg/l).

PERSENTASE PENETASAN TELUR IKAN BETOK (Anabas testudineus)  DENGAN SUHU INKUBASI YANG BERBEDA

ABSTRACT

The aims of this study was to know the effect of different temperatures on the percentage of climbing perch  eggs hatching. This study has been done from October 2012 to November 2012 at Fish Breeding Unit “Batanghari Sembilan” Indralaya. The implementation of this study include selection of the brood, rearing of brood stock, injection ovaprim, spawning, hatching eggs, larval rearing. The results  indicate that the most rapid hatching time obtained in P4 treatment (34° C) with a incubation time 964 minutes. Optimal incubation temperature to produce the maximum hatching percentage was 98.66% at a temperature of 34 oC. Water quality of medium were 31-340C for temperature, pH  6,7-7,6 and disolved oxygen 3,3-3,8 mg.L-1.

ABSTRACT

The aim of this research is to determine the chromosomes number of snakehead fish (Channa striata) from floodplain area, highland swamp, and tidal land. The research has done on December 2013 until January 2014 at Laboratorium Pengembangbiakan dan Genetika Ikan, Department of Aquaculture, Faculty of Fishries and Marine Science, Bogor Agricultural Institute, Laboratorium Dasar Perikanan dan Laboratorium Budidaya Perairan, Study Program of Aquaculture, Agriculture Faculty, Sriwijaya  University and Laboratorium Fisiologis Hewan, Mathematics and Natural Science Faculty, Sriwijaya University, Indralaya. The research was conducted by means of direct observation and analyzed descriptive through making preparations chromosome squash methods. The results showed no difference in diploid (2n) chromosome number of snakehead fish floodplain area as much ranges 40-42, highland swamps ranges 36-40 and tidal land ranges 38-40 and it can be concluded that the chromosomes amount of snakehead fish  between 36-42.

ABSTRACT

The aimed of this research was to find out survival rate and growth of kissing
gouramy larvae with different stocking density. Research used Completely Randomized
Design (CRD) with five treatments and three replications. The treatment was used
different of stocking density were 6, 8, 10, 12, and 14 larvae per liter. Result of research
showed that the different of stocking density was significant on length and weight
growth, but not significant for survival rate. Stocking density with six larvae per liter
was resulted the highest value consist of absolute length growth (3.2±0.0 cm per larvae),
absolute weight growth (476.67±11.55 mg per larvae) and survival rate (60.55±1.27 %).
Physical-Chemical parameters of water during research was in optimal range were temperature 26-280C, pH 6.9-7.1, dissolved oxygen 4.00-4.09 mg.L-1 and ammonia 0.004–0.013 mg.L-1.

ABSTRACT
The aim  of the research was to fiad out sex ratio proportion on ovulation time, fecundity, fertilization, and hatching percentage of climbing perch eggs. The research used Completely Randomized Design with four treatments and three replicates with P as treatment code. The treatment used was ratio between female and male including P1(1:1), P2(1:2), P3(1:3), and P4(1:4). Observed parameters were ovulation time, fecundity, fertilization, hatching percentage, and water quality. Observed parameters were ovulation time, fecundity, fertilization, hatching percentage, and water quality. The Results showed that all treatment did not affected significantly affentedthe observed parameter (P<0,05). P2 treatment provided the fastest ovulation time with 137.33 minute, highest fecundity value provided by P4 with 18.533 eggs grains. Eggs fertilization rates of eggs on each treatmentwas 100%, and the highest hatching percentage was given by P1 treatment withnumber 91.57%. Ranges of water quality parameters recorded on  research   were temperature 28-29 0C, pH 6.2-6.5, disollved oxygen 6.11-6.79 ppm and  amonia 0.010-0.038 ppm.

ABSTRACT

The purpose of this research were to determine the best ratio of injection pituitary extract of climbing perch, latency time, amount of egg,  percentage fertilized of egg, percentage hatching of egg and survival rate of climbing perch pro larvae. The research was conducted at Laboratorium Dasar Perikanan, Department of Aquaculture, Agriculture Faculty, Sriwijaya University since September until November 2014. The research used completely randomized design with 4 treatments and 3 replications. Those were P1 (1 donors : 1 recipient), P2 (2 donors: 1 recipient), P3 (3 donors : 1 recipient) and K(+) (®ovaprim 0.5 ml/kg). Based on to the honest significant different test showed that climbing perch pituitary extract injection of different ratio of weight fish had significantly different to latency time, amount of egg rate, percentage fertilized of egg but it’s not significantly different to percentage hatching of egg and survival rate. The best spawning latency time was P3 (9 hours 25 minutes), amount of egg was K(+) (10,123 grain), the highest percentage fertilized of eggs was P3 (98.25 %), the highest hatching percentage of eggs was P2 (93.90 %) and the highest survival rate pro larvae of climbing perch was P1 (87.71 %).

ABSTRACT

The climbing perch (Anabas testudineus) is very difficult to spawn naturally in the cultivation environment. The aim of this research was to know the best dose of broiler pituitary extract for latency period, fecundity, fertilized egg percentage, hatching egg percentage, survival rate of climbing perch (D1-D3) larvae. This research had been conducted from April-May 2015 at Laboratory of Budidaya Perairan,  Aquaculture Department, Faculty of Agriculture, Sriwijaya Unversity, Indralaya. This research method used completely randomized design (CRD) with four treatments and three replications. Females broodstock were induced by broiler pituitary extract with different doses that were, 400 mg/kg, 500 mg/kg, 600 mg/kg body weight and Induced by that synthetic gonadotrophin hormone (positif control) 0,5 ml/kg body weight. The parameters observed during the research were latency period, fecundity, fertilized egg percentage, hatching egg percentage, survival rate of climbing perch (D1-D3) larvae and water qualities. The result of this research showed that if considered from latency period and fecundity that synthetic gonadotrophin hormone more effective compared with broiler pituitary extract, but if in terms of the fertilized egg percentage, hatching egg percentage, and survival rate of climbing perch (D1-D3) larvae, that synthetic gonadotrophin hormone as effective as broiler pitutary extract. The range of water qualities were temperature 28-30 0C, pH 5.34-6.98, DO 3.135.97 ppm, and ammonia 0.009- 0.0018 ppm.

ABSTRACT

The aims of this research was to study Gonad Maturity Level (GML), Index Gonado Somatik (IGS), absolute fekundity and egg diameter of Channa striata injected with the Human Chorionic Gonadotropin hormone. The research was conducted in 26th of December 2013 until 26th of January 2014 at the Fish Hatchery Unit (FHU) Batanghari Sembilan, North Indralaya District, Ogan Ilir, South Sumatra. Channa striata which as used had weights 85-105 gram per fish. The experiment was arranged in a Completely Randomized Design (CRD), (P0) No injection of HCG, (P1) 200 IU per kg fish, (P2) 250 IU per kg fish, (P3) 300 IU per kg fish. Parameters observed Gonad Maturity Level (GML), Index Gonado Somatik (IGS), absolute fekundity and egg diameter. From the four treatment experiment, P3 treatment produced the highest GML V whose the percentage was 20%, while P0 treatment did not reach GML IV and V, most of the P0 treatment (80%) only reached GML II. Based on further test of Duncan Signipicant Differences with 95% credibility level, the IGS data of Channa striata was significantly different and influential among treatments, the smallest IGS was produced from P0 treatment that is 0.43% while the largest IGS was produced from P3 treatment that is 5.91%. The smallest absolute fecundity was on the P0 treatment which was 582 eggs while the largest was on P3 treatment that is 5,775 eggs. The smallest egg diameter was on the P0 treatment which was 0.35 mm while the largest was on P3 treatment that is 0.93 mm. Hormone HCG injections of 300 IU per kg weight of the fish proved to be finalizing a Channa striata gonad.

MASKULINISASI IKAN GAPI (Poecilia reticulata) MELALUI PERENDAMAN INDUK BUNTING DALAM LARUTAN MADU DENGAN LAMA  PERENDAMAN BERBEDA


ABSTRACT
The purpose of this research was to know the influence of dipping time of pregnant guppy in 5 ml.L-1 of honey solution to masculinitation of guppy. The research was conducted on July until October 2012 at Fish Breeding Unit of Batanghari Sembilan Foundation, Indralaya, Ogan Ilir. Research conducted using completely randomized design with four treatments of dipping namely P0 (0 hours), P1 (12 hours), P2 (14 hours), and P3 (16 hours) with three replications. The parameters observed were male percentage, survival rate, and water quality. The result of the current research showed that the dipping time of pregnant guppy in 5 ml.L-1 honey solution gave significant influence on male percentage of guppy larva. The percentages of male guppy from highest to lowest were P0 (69,26%), P1 (76,66%), P2 (52,23%), and P3 (40%). The dipping time of pregnant guppy in 5 ml.L-1 honey solution gave not significant influence on survival rate of guppy larva. Water quality is still in range appropriate to survival rate of guppy

ABSTRACT

The purpose of this study was to determine the effect of different incubation temperatures to the rate of the kissing gouramy (Helostoma temminckiiC.V) yolk absorption, and to know the right time to start feeding in the larval stage. This study was held on January 09th to 13th, 2013 using Completely Randomized Design (CRD) with five treatments namely P1 (26±0.50C), P2 (28±0.50C), P3 (30±0.50C), P4 (32±0.50C) and P5 (34±0.50C).Each treatment was repeated three times. The results showed that the temperature gave significant effect (P>0,05) to rate of yolk absorption, periode of yolk absorption and percentage of larval survival.The highest rate of yolk absorption was P5 (42.24 x 10-5 mm3/hour), but not significantly different fromP4 (41.63 x 10-5 mm3hour-1), and P3 (41.83 x 10-5 mm3hour-1). The fastest periode of yolk absorption was P5 (72 hours) and the longest was P1 (78.67 hours). While the highest larval survival rate was found on P3 (89.87%), on the other hand the lowest survival rate was found on P5 (62.35%). The natural feeding of kissing gouramy fish should be started at 74th hours (3 days, 2 hours) after hatching and larval rearing should use temperature of 300 ± 0.50 C.

ABSTRACT
The Problems of climbing perch hatchery are low survival and growth rate in larvae. This is associated with the larvae ability to receive feed transition from the endogenous feeding to the exogenous feeding. During this period, feed must be consistent with required by the larvae either natural feed, artificial feed, or a combination. Naturaly silk worms is given, however availability silk worms is very limited in the wild. There to need to combine silk worms with artificial feed. The purpose of this study is to determine growth and survival rate of climbing perch larvae which is fed silk worms combined with artificial feed. The research had been conducted in Laboratorium Dasar Perikanan, Department of Aquaculture, Agriculture Faculty, Sriwijaya University on January until March 2016. The research method used a completely randomized design with five treatments and three replications. The treatments were P1 (silk worms 100% + 0% artificial feed), P2 (silk worms 75% + 25% artificial feed), P3 (silk worms 50% + artificial feed 50%), P4 (silk worms 25% + 75% artificial feed) and P5 (silk worms 0% + 100% artificial feed). Parameters that had been observed were survival rate, specific growth rate, and water quality parameters. The results showed that culture of climbing perch larvae fed silk worms, artificial feed and combination of them gave significant effect on survival rate and specific growth rate (weight and length). The treatment P3 resulted highest of survival rate 88.67% and specific growth rate (weights 20.16%.days-1 and length 8.11%.days-1). During the  research, water quality were in tolerance range for climbing perch larvae  rearing: temperature was  27-29 oC, pH 6.2-7.3, DO 4.11-5.50 mg.L-1, ammonia 0.01-0.21 mg.L-1.
